At a shell prompt if you type:  cat xsetup
What is the mouse option? You can edit xsetup and change the mouse options. The options for the mouse are:

   -mouse path,n

Where path would be /dev/ttyS0
and n is the number of buttons.

I don't have a serial mouse, so this is untested. Good Luck.
